Cytotoxicity and mycotoxin production of shellfish-derived Penicillium spp., a risk for shellfish consumers ArchiMer
Geiger, Marie; Guitton, Yann; Vansteelandt, M.; Kerzaon, I.; Blanchet, Estelle; Du Pont, T. Robiou; Frisvad, J. C.; Hess, Philipp; Pouchus, Y. F.; Grovel, Olivier.
In order to assess the putative toxigenic risk associated with the presence of fungal strains in shellfish-farming areas, Penicillium strains were isolated from bivalve molluscs and from the surrounding environment, and the influence of the sample origin on the cytotoxicity of the extracts was evaluated. Extracts obtained from shellfish-derived Penicillia exhibited higher cytotoxicity than the others. Ten of these strains were grown on various media including a medium based on mussel extract (Mytilus edulis), mussel flesh-based medium (MES), to study the influence of the mussel flesh on the production of cytotoxic compounds. The MES host-derived medium was created substituting the yeast extract of YES medium by an aqueous extract of mussel tissues, with...

Évaluation d’une suite de bio-essais pour la détection et l’étude de composés lipophiles de micro-organismes marins issus de mollusques bivalves et de leur environnement ArchiMer
Geiger, Marie.
The context of this study was the assessment of the safety of edible bivalve molluscs. Bivalve can indeed accumulate high concentrations of phycotoxins into their hepatopancreas, potentially leading to human intoxication outbreaks after consumption of such contaminated bivalves. In order to ensure the protection of shellfish consumers, regulated toxins are monitored using physico-chemical techniques. In parallel, emerging toxins, that can be putatively produced by microalgae or marine fungi, are monitored using the intraperitoneal mouse bioassay. However, this bioassay suffers ethical and methodological drawbacks. Thus, the objective of this work was to assess the ability of a bioassay suite to detect lipophilic toxins. This suite was composed of three...

Integrated control of Penicillium digitatum by the predacious yeast Saccharomycopsis crataegensis and sodium bicarbonate on oranges BJM
Pimenta,R. S.; Silva,J. F. M.; Coelho,C. M.; Morais,P. B.; Rosa,C. A.; Corrêa Jr,A..
Our investigation of integrated biological control (IBC) started with an assay testing activity of the predacious yeast Saccharomycopsis crataegensis UFMG-DC19.2 against Penicillium digitatum LCP 4354, a very aggressive fungus that causes postharvest decay in oranges. Under unfavourable environmental conditions, the yeast showed a high potential for control (39.9% disease severity reduction) of this fungus. This result was decisive for the next step, in which S. crataegensis was tested in association with sodium bicarbonate salt, a generally regarded as safe (GRAS) substance. The yeast was able to survive at different concentrations of the salt (1%, 2% and 5%), and continued to grow for a week at the wound site, remaining viable at high population for 14...

Isolation of recombinant strains with enhanced pectinase production by protoplast fusion between Penicillium expansum and Penicillium griseoroseum BJM
Varavallo,Maurilio Antonio; Queiroz,Marisa Vieira de; Lana,Taís Guimarães; Brito,Admilson Toscano Ribeiro de; Gonçalves,Daniel Bonoto; Araújo,Elza Fernandes de.
Protoplast fusion between complementary auxotrophic and morphological mutant strains of Penicillium griseoroseum and P. expansum was induced by polyethylene glycol and calcium ions (Ca2+). Fusant strains were obtained in minimal medium and a prototrophic strain, possibly diploid, was chosen for haplodization with the fungicide benomyl. Different recombinant strains were isolated and characterized for occurrence of auxotrophic mutations and pectinolytic enzyme production. The fusant prototrophic did not present higher pectinase production than the parental strains, but among 29 recombinants analyzed, four presented enhanced enzyme activities. The recombinant RGE27, which possesses the same auxotrophic and morphologic mutations as the P. griseoroseum...

Novel antimicrobial secondary metabolites from a Penicillium sp. isolated from Brazilian cerrado soil Electron. J. Biotechnol.
Petit,Philippe; Lucas,Esther M. F.; Abreu,Lucas M; Pfenning,Ludwig H; Takahashi,Jacqueline A.
The morphological features of a Penicillium, isolated from Brazilian cerrado soil, were characterized and showed to be distinctly different from all well-defined Penicillium species. Chemical and biological investigation on the ethyl acetate extract of this Penicillium isolate resulted in the isolation of three new naphthalenoids: a major metabolite, methyl 6-acetyl-4-methoxy-5,7,8-trihydroxynaphthalene-2-carboxylate and two minor ones, methyl 6-acetyl-4-methoxy-7,8-dihydroxynaphthalene-2-carboxylate and methyl 6-acetyl-4-methoxy-5,8-dihydroxynaphthalene-2-carboxylate. Their structures were determined based on their mono and bidimensional nuclear magnetic resonance data. Acetyl, allyl and methoxyl derivatives of the major metabolite were prepared in order...

Production of ß-Glucosidase by Penicillium purpurogenum BJM
Dhake,A.B.; Patil,M.B..
The fungus Penicillium purpurogenum was found to produce intracellular ß-glucosidase. Maximum activity of ß-glucosidase was observed on sucrose. Various cultural parameters of cultivation of P. purpurogenum for production of ß-glucosidase were optimized. Maximum enzyme content was observed after 96 hours of cultivation at 30º;C. Addition of amino acids histidine and cysteine induced ß-glucosidase synthesis to certain extent. The optimum temperature and pH for ß-glucosidase activity was 50º;C and 5.5 respectively. ß-glucosidase of P.purpurogenum shows stability at pH 2 thus it could be an ideal enzyme for debittering in fruit juice and wine industries.
